Human relaxin-2 (RLN2) is a peptide hormone best known for its role in female reproduction, which softens the cervix and pubic ligaments to prepare for childbirthIt is a 6 kDa, heterodimeric protein composed of an A-chain and a B-chain that circulates in the blood, where it interacts with receptors to affect various tissues. Beyond reproduction, it is involved in other physiological processes and may be a therapeutic target for conditions like heart failure and fibrosis. 

Structure and function
    • Structure

      RLN2 is a 6 kDa protein made of two polypeptide chains: a 24-amino acid A-chain and a 29-amino acid B-chain, linked by three disulfide bonds. It is structurally related to insulin. 

  • Reproductive role

    It is most famous for its role in pregnancy, where it causes the relaxation of the pubic symphysis and the softening of the cervix. 

  • Mechanism
    RLN2 exerts its effects by binding to specific receptors, primarily LGR7 and LGR8, which are members of the relaxin family peptide (RXFP) receptors. This binding activates various signaling pathways that influence tissue remodeling and other cellular processes.
